ABSTRACT

Plastination is a technique used to preserve biological tissues while retaining most of their original appearance. In the technique, developed by Dr. Gunther von Hagens in 1977, specimens were impregnated with a polymer, such as silicone, epoxy, or polyester. Considered the most suitable material for brain plastination, polyester has a wide application in teaching and research compared with imaging techniques. The materials for plastination are usually imported from Germany and more expensive than domestic products. If domestic polymers were to enter the market it would favor the expansion of plastination in Brazil. Hence, this study evaluated the feasibility of using domestic polyesters to replace the usual Biodur® (P40) in plastination of brain slices. For this evaluation, 2-mm-thick sections of bovine brains were prepared and plastinated with domestic polyester. Slices were compared before impregnation and after curing using standardized photographs taken after dehydration and after curing. Plastination followed the standard protocol: fixation, dehydration, forced impregnation, and curing. Fifteen brain slices were plastinated with each polyester (P40, P18, and C1-3). There was no significant difference in the percent shrinkage between groups after plastination of P18 and P40, but the curing time of Cristalan© polymer was too short for impregnation. Therefore, no initiator was used for C polymers impregnation. Thus, domestic polyester P18 was a viable option for the process.

ABSTRACT

Resumo Fundamento Embora os ácidos graxos poli-insaturados ômega-3 e ômega-6 (AGPIs n-3 e n-6) tenham efeitos bem conhecidos sobre os fatores de risco de doenças cardiovasculares (DCV), ainda existe um conhecimento limitado sobre como eles afetam os indicadores de qualidade da LDL. Objetivo Avaliar as associações dos AGPIs n-3 e n-6 de hemácias com o tamanho da partícula da LDL, LDL-c pequena e densa (sdLDL-c) e com LDL eletronegativa [LDL(-)] em adultos com fatores de risco para DCV. Métodos Estudo transversal com 335 homens e mulheres de 30 a 74 anos com, pelo menos, um fator de risco cardiovascular. Foram realizadas análises de parâmetros bioquímicos, como glicose, insulina, HbA1c, proteína C reativa (PCR), perfil lipídico, subfrações de lipoproteínas, partícula eletronegativa de LDL [LDL(-)] e seu autoanticorpo, e os AGPIs n-3 e n- 6 de hemácias. Os testes t independente/teste de Mann-Whitney, ANOVA unidirecional/teste de Kruskal-Wallis e regressões lineares múltiplas foram aplicados. Todos os testes foram bilaterais e um valor de p inferior a 0,05 foi considerado estatisticamente significativo. Resultados A relação n-6/n-3 de hemácias foi associada ao aumento dos níveis de LDL(-) (β = 4,064; IC de 95% = 1,381 - 6,748) e sdLDL-c (β = 1,905; IC de 95% = 0,863 - 2,947), e redução do tamanho das partículas de LDL (β = -1,032; IC de 95% = -1,585 − -0,478). Individualmente, os AGPIs n-6 e n-3 apresentaram associações opostas com esses parâmetros, realçando os efeitos protetores do n-3 e evidenciando os possíveis efeitos adversos do n-6 na qualidade das partículas de LDL. Conclusão O AGPI n-6, presente nas hemácias, foi associado ao aumento do risco cardiometabólico e à aterogenicidade das partículas de LDL, enquanto o AGPI n-3 foi associado a melhores parâmetros cardiometabólicos e à qualidade das partículas de LDL.


Abstract Background While Omega-3 and omega-6 polyunsaturated fatty acids (n-3 and n-6 PUFAs) have established effects on cardiovascular disease (CVD) risk factors, little is known about their impacts on LDL quality markers. Objective To assess the associations of n-3 and n-6 PUFA within red blood cells (RBC) with LDL particle size, small dense LDL-c (sdLDL-c), and electronegative LDL [LDL(-)] in adults with CVD risk factors. Methods Cross-sectional study involving 335 men and women aged 30 to 74 with at least one cardiovascular risk factor. Analyses were conducted on biochemical parameters, such as glucose, insulin, HbA1c, C-reactive protein (CRP), lipid profile, lipoprotein subfractions, electronegative LDL particle [LDL(-)] and its autoantibody, and RBC n-3 and n-6 PUFAs. Independent t-test/Mann-Whitney test, one-way ANOVA/Kruskal-Wallis test, and multiple linear regressions were applied. All tests were two-sided, and a p-value of less than 0.05 was considered statistically significant. Results The RBC n-6/n-3 ratio was associated with increased LDL(-) (β = 4.064; 95% CI = 1.381 - 6.748) and sdLDL-c (β = 1.905; 95% CI = 0.863 - 2.947) levels, and reduced LDL particle size (β = -1.032; 95% CI = -1.585 − -0.478). Separately, n-6 and n-3 PUFAs had opposing associations with those parameters, reinforcing the protective effects of n-3 and showing the potential negative effects of n-6 on LDL particle quality. Conclusion RBC n-6 PUFA was associated with increased cardiometabolic risk and atherogenicity of LDL particles, while n-3 PUFA was associated with better cardiometabolic parameters and LDL particle quality.

ABSTRACT

The abnormal lipids metabolism is a critical pathological feature of coronary heart disease (CHD). Additional supplemental intake of polyunsaturated fatty acid (PUFA) has long been considered to be an effective strategy for preventing CHD, but more and more clinical trials have denied this view. Still, it is ambiguity for the specific mechanism of PUFA in CHD. The experimental programs are compliant with ethical principles for animal use and have been approved by the Animal Experiment Ethics Committee of Jinan University. In the present study, we established an animal model by intake of omega-6 PUFA combined acute myocardial ischemia to explore the mechanism of CHD. Intragastric administration of linoleic acid (LA) for 14 days, intraperitoneal injection of isoprenaline (ISO) was applied to induce acute myocardial ischemia for the animal model establishment. The animal ultrasound imaging system was used to detect cardiac function in vivo after ISO injection for 24 h. Serum and heart tissue samples were collected for the myocardial enzyme, phospholipidomics analysis and molecular biological detection. Compared to the LA group, the cardiac function showed that the left ventricular ejection fraction (EF%) and the left ventricular shortening fraction (FS%) decreased, aspaetate aminotransferase (AST), creatine kinase isoenzyme (CK-MB), and lactate dehydrogenase (LDH) increased in the LA + ISO mice. Compared to the ISO group, the phospholipidomics analysis showed that the PUFAs significantly were raised in the LA + ISO myocardium, and the content of oxidized phosphatidylethanolamine (ox-PE) changed most remarkable. Compared with the ISO group, the molecular biology detection showed that glutathione (GSH) and nicotinamide adenine dinucleotide phosphate (NADPH) were depleted, the end-products of ox-PE were increased, and the level of arachidonic acid 12/15-lipoxygenase (ALOX15) protein expression increased obviously. We suggest that ALOX15 mediated phospholipid peroxidation might be the critical mechanism of LA increased the susceptibility of myocardial ischemia injury. This study provides an experimental basis for whether PUFA could be used as an alternative treatment strategy for CHD prevention and provides a new intervention target for the early prevention strategy of CHD.

ABSTRACT

Abstract The present study has been carried out with the seed extracts of Nyctanthes arbor-tristis L. (Parijat) and evaluates its antioxidant potential and profiling the phytochemical constituents by Gas Chromatography-Mass Spectrometry (GC-MS) analysis. The antioxidant potential of the seed extracts was measured by four different in vitro assay like 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, superoxide anion free radical scavenging activity, ferric reducing antioxidant power (FRAP) and lipid peroxidation inhibition potential (LPIP) assay. The total phenol content (TPC) and total flavonoid content (TFC) were estimated. The ethyl acetate extract (EAE) of seeds showed potential DPPH free radical scavenging activity (EC50 129.49±3.55µg/ml), superoxide anion radical (EC50 969.94±8.03µg/ml) and LPIP (EC50 452.43±5.07 µg/ml) activities. The total phenol content was maximum in aqueous extract (AQE) which was 201.00±0.20 µg/mg gallic acid equivalent. The EAE was rich with total flavonoid and it was found to be 34.50±0.40 µg/mg rutin equivalent. The EAE was subjected for phytochemical-profiling using GC-MS system. The presence of different phytoconstituents supports the medicinal value of the seeds. The results suggest that EAE constitutes a promising new source of novel compounds. Further, it can be used for isolation and purification of specific compounds which have good antioxidant activities and possess useful biological activities.

ABSTRACT

RESUMEN Obtener la envolvente de ruptura de suelos no saturados requiere de equipos especializados, de elevado costo y difícil acceso. Sin embargo, a través de ensayos de laboratorio simples combinados como corte directo, tracción indirecta y succión, es posible determinar la envolvente. La presente investigación busca, por tanto, validar dicha metodología, en un material limo de origen residual de la ciudad de Medellín-Antioquia, Colombia, compactado a la máxima densidad a través del ensayo Proctor normal. La validación se efectúa por medio de la repetitividad de una serie de ensayos de corte directo para diferentes valores de succión; comprobando que, es posible encontrar la envolvente de ruptura al corte de un suelo fino residual compactado, para distintas condiciones de succión, por medio de los ensayos de laboratorio de corte directo en condición consolidada y drenada, succión por el método del papel filtro y tracción indirecta, cuando no se cuenta con equipos de corte con succión controlada.


ABSTRACT Obtaining the failure envelope of unsaturated soils requires specialized equipment, which is expensive and difficult to access. Nevertheless, by combining simple laboratory tests like direct shear test on saturated samples, indirect tension test, and soil suction measurements by filter paper it is also possible to determine the envelope. This research aims to validate this latter methodology on a residual origin silt material from the city of Medellin, Colombia, which is compacted to the maximum dry density through the standard proctor test. The validation is carried out by repeating a series of direct shear tests for different suction values. It was verified that it is possible to find the shear failure envelope of a compacted fine-grained residual soil for different suction conditions, using the direct shear test under consolidated drained conditions, suction by the filter paper method, and the indirect tension test, when no suction controlled shear equipment is available.


RESUMO A obtenção da envoltória de ruptura de solos não saturados com controle de sucção requer equipamentos especiais, que são caros e de difícil acesso. No entanto, através de ensaios simples laboratoriais combinados, tais como os de cisalhamento direto, tração indireta e sucção, é possível determinar a envoltória de resistência. A presente investigação procura validar esta metodologia, usando um material siltoso de origem residual da cidade de Medellin-Antioquia, Colômbia, compactado na condição de densidade máxima obtida no ensaio de compactação Proctor normal. A validação é feita por meio da realização de uma série de ensaios de cisalhamento direto para diferentes valores de sucção; mostrando que é possível encontrar a envoltória de ruptura de um solo fino residual compactado, para diferentes condições de sucção, por meio de ensaios laboratoriais de cisalhamento direto consolidados drenados, sucção determinada pelo método do papel de filtro e ensaio de tração indireta, quando não se dispõe de equipamento de cisalhamento direto à sucção controlada.

RESUMEN La etapa postprandial se ha relacionado con cambios en marcadores inflamatorios, bioquímicos y celulares. El objetivo de este estudio fue evaluar los efectos postprandiales del agregado de aceite de Sacha inchi sobre la concentración y tamaño de leucocitos, eritrocitos y plaquetas, y en marcadores de inflamación, después de la ingesta de una comida rica en grasas. Una muestra de 42 individuos aparentemente sanos de sexo masculino consumió dos desayunos, uno de ellos adicionado con 15 mL de aceite rico en ácidos grasos poliinsaturados extraído de semillas de Sacha Inchi. Se tomaron muestras de sangre en ayunas y a las 4 horas postprandiales para determinar variables hematológicas (número y tamaño de leucocitos y plaquetas y distribución porcentual de leucocitos) y bioquímicas como interleukina 6 (IL6) y proteína C reactiva de alta sensibilidad (PCR-as). Con ambos desayunos aumentaron la concentración de PCR y los recuentos de leucocitos y plaquetas. El agregado de aceite de Sacha inchi aumentó el porcentaje de linfocitos (p= 0,005) y disminuyó el de granulocitos (p= 0,012), revirtiendo el aumento de la relación evidenciada luego de la ingesta grasa y las concentraciones de IL6. Los resultados permiten concluir que el agregado de 15 mL de aceite de Sacha inchi a un desayuno rico en grasas afecta la relación entre las diferentes poblaciones leucocitarias, lo que podría atenuar los efectos inflamatorios postprandiales y el riesgo cardiovascular. ABSTRACT The postprandial stage is related to an increase in biochemical and cellular inflammatory markers. The objective of this study was to evaluate the postprandial effects of the addition of Sacha inchi oil on the concentration and size of leukocytes, erythrocytes and platelets, and on inflammation markers, after the ingestion of a high-fat meal. A sample of 42 seemingly healthy male individuals consumed two high-fat breakfast meals, one with the addition of 15mL of oil rich in polyunsaturated fatty acids (omega 3 and 6 series), extracted from Sacha Inchi seeds. Fasting blood samples were taken at 4 hours postprandial to determine hematological variables (number and size of leukocytes and platelets and percentage distribution of leukocytes) and biochemical variables such as interleukin 6 (IL6) and high-sensitivity C-Reactive protein (hs-CRP). The concentration of CRP and leukocyte and platelet counts increased following ingestion of both types of breakfast. The addition of Sacha inchi oil increased the percentage of lymphocytes (p= 0.005) and decreased that of granulocytes (p= 0.012), reversing the increase in the granulocytes / lymphocytes ratio evidenced after fat intake. The percentage of intermediate-sized cells and postprandial concentrations of IL6 also decreased. In conclusion, the addition of 15.0 mL of Sacha inchi oil to a high-fat breakfast modulates the relationship between different leukocyte populations, which could mitigate postprandial inflammatory effects and cardiovascular risk. ABSTRACT

One-sixth of the currently known natural products contain α, β-unsaturated carbonyl groups. Our previous studies reported a rare C-sulfonate metabolic pathway. Sulfonate groups were linked to the β-carbon of α, β-unsaturated carbonyl-based natural compounds through this pathway. However, the mechanism of this type of metabolism is still not fully understood, especially whether it is formed through enzyme-mediated biotransformation or direct sulfite addition. In this work, the enzyme-mediated and non-enzymatic pathways were studied. First, the sulfite content in rat intestine was determined by LC-MS/MS. The results showed that the amount of sulfite in rat intestinal contents was from 41.5 to 383 μg·g

ABSTRACT

This study evaluated the fatty acid composition and qualitative characteristics of meat from lambs feed in feedlot with increasing levels of canola grain. Meat obtained from 27 lambs fed 0, 8 and 16% inclusion of canola grain were analyzed. There was a reduction in the content of saturated fatty acids (SFA): lauric, heptadecanoic, and stearic; and unsaturated fatty acids (UFA): palmitoleic, oleic, and eicosatrienoic, with the increasing levels of canola grain. There was a linear reduction for the ω3 acid and the ω3: ω6 ratio, while the ω6: ω3 ratio increased. But the values observed for this ratio (ω6: ω3) were lower than 4, which is considered a satisfactory value. There was a slight increase (~2%) for the thrombogenicity index and atherogenicity index with the inclusion of canola grain. There was a reduction in the hypocholesterolemic: hypercholesterolemic ratio, with mean values of 2.09 for the diet with 0% inclusion, and 2.06 for 8 and 16% inclusion of canola. Although the inclusion of canola grain for lambs decreased the content of some UFA, reflecting alterations in the correlated nutritional properties, there is a reduction of SFA. The results for instrumental analysis, proximate composition, and sensory acceptance of the lamb meat were similar among the treatments.(AU)


Objetivou-se avaliar a composição dos ácidos graxos e as características qualitativas da carne de cordeiros terminados com níveis crescentes de canola grão na dieta. Foram estudadas amostras de carne provenientes de 27 cordeiros, confinados por 45 dias e alimentados com 00, 08 e 16% de inclusão de canola grão. Houve efeito para os ácidos graxos saturados (AGS): laurico, heptadecanoico e esteárico; ácidos graxos monoinsaturados (AGMI): palmitoleico e oleico; e ácidos graxos polinsaturados (AGPI): eicosatrienoico. Estes ácidos reduziram à medida que os níveis da canola grão aumentaram. Para os AGMI, houve efeito com maior concentração no músculo dos cordeiros alimentados sem a canola grão (49,80%). Houve uma redução linear para o ácido ω3 e para a relação ω3:ω6, enquanto que a relação ω6:ω3 aumentou. A canola grão influenciou o índice de aterogenicidade, com média de 0,57 para o tratamento com 0% de inclusão e 0,58 para os tratamentos com 8% e 16% de inclusão. Observou-se aumento linear para o índice de trombogenicidade com a inclusão da canola grão, cujas médias foram, respectivamente, 0,16, 0,17 e 0,18 para os tratamentos 0%, 8% e 16% de inclusão. Houve redução para a relação hipocolesterolêmicos-hipercolesterolêmicos, com médias de 2,09 para a dieta com 0% de inclusão, e 2,06 para 8% e 16% de inclusão da canola em grão na dieta. A canola grão não influenciou a análise instrumental, composição centesimal e atributos sensoriais da carne.(AU)

ABSTRACT

In the present study a series of 34 synthetic ligustrazine-containing α, ß-Unsaturated carbonyl-based compounds and oximes, recognized as anticancer compounds were assessed against protozoa of the Trypanosoma and Leishmania species. Ligustrazine, chemically known as tetramethylpyrazine (TMP), was selected as the core moiety for the synthesis of α, ß-Unsaturated carbonyl-based compounds and these compounds were selected as precursors for the synthesis of new oximes. Some derivates, including 5f and 6i, showed multiple activities against all tested strains. In particular compounds 5f and 8o are the most potent and they are, therefore, potential candidates for trypanosomiasis and leishmaniasis

ABSTRACT

RESUMEN El envejecimiento en general, y el cutáneo, en particular, es un proceso deletéreo y universal. El envejecimiento cutáneo se puede dividir en envejecimiento cronológico y fotoenvejecimiento, este último se activa a través del daño en la piel humana, atribuible a la exposición repetida a los rayos ultravioleta (UV) de la luz solar. Se ha considerado que los ácidos grasos derivados del aceite de pescado (omega 3) están asociados con la fotoprotección de la piel. El objetivo de este trabajo es describir los efectos del omega 3 como medicamento antienvejecimiento en la población de adultos mayores. Para ello, se realizó una revisión bibliográfica de artículos publicados entre enero de 2010 y junio de 2020, en Pubmed y Google Scholar. La literatura disponible evidencia el efecto y beneficio de estos ácidos grasos en el proceso de envejecimiento, los cuales constituyen terapias clínicas a nivel dermatológico y en áreas de cirugía plástica. Su uso ejerce un efecto positivo sobre el proceso de envejecimiento en diferentes órganos, mediante la acción frente al estrés oxidativo.


ABSTRACT Aging in general, and skin aging in particular, is a deleterious and universal process. Skin aging can be divided into chronological aging and photoaging, the latter being activated through damage to human skin, attributable to repeated exposure to ultraviolet (UV) rays from sunlight. Fatty acids derived from fish oil (omega 3) have been considered to be associated with photoprotection of the skin. The objective of this work is to describe the effects of omega 3 as an anti-aging drug in the elderly population. A bibliographic review of articles published between January 2010 and June 2020, in Pubmed and Google Scholar, was carried out. The available literature shows the effect and benefit of these fatty acids in the aging process, which constitute clinical therapies at the dermatological level and in areas of plastic surgery. Its use has a positive effect on the aging process in different organs, through action against oxidative stress.

ABSTRACT

RESUMEN El contenido de ácidos grasos en la leche materna madura varía dependiendo del estado nutricional de la madre, de la dieta materna antes de la gestación y durante la gestación, el tiempo de lactancia, entre otros. Por lo anterior, este estudio pretende caracterizar la ingesta dietaría de madres donantes de leche materna madura y determinar el perfil de ácidos grasos con énfasis en los ácidos grasos poliinsaturados de cadena larga. Para ello, se contó con la participación de 50 madres lactantes y donantes las cuales se extrajeron cada una, 60 ml de leche materna madura, para determinar el perfil de ácidos grasos por cromatografía de gases. También, se determinó el consumo de ácidos grasos por frecuencia de consumo y por consumo habitual. Se reporto que en la leche materna madura prima el contenido de ácidos grasos insaturados (62%) sobre los saturados (38%), además, el total de grasa encontrado, fue superior al reportado normalmente en la literatura. Finalmente, no se evidencio correlación entre el consumo de ácidos grasos en la dieta de las madres y el contenido de estos en la leche materna madura, aspecto coherente con el contenido nutricional adecuado que debe aportar este alimento al lactante sin afectar su salud.


ABSTRACT The content of fatty acids in mature breast milk varies depending on the nutritional status of the mother, maternal diet before pregnancy and during pregnancy, and breastfeeding time, among others. This study aimed to characterize the dietary intake of donor mothers of mature breast milk and determine fatty acid profile with emphasis on long-chain polyunsaturated fatty acids. Fifty donor mothers were involved, each one provided 60 ml of mature breast milk; fatty acid profile was determined by gas chromatography. The consumption of fatty acids was determined by frequency of consumption and by habitual semiquantitative consumption. We found that unsaturated fatty acid content was greater than saturated (62%) in mature breast milk and total fat was higher than that normally reported in the literature. Finally, there was no correlation between the consumption of fatty acids in the diet of the mothers and the content of these in mature breast milk, which is consistent with the adequate nutritional content that this food should provide to the infant without affecting their health.

ABSTRACT

Introducción: la diabetes tipo 2 es una enfermedad cuya prevalencia se incrementa rápidamente en los países de ingresos medianos y bajos. Siendo los carbohidratos el principal determinante de la glucemia posprandial, se ha evidenciado que existe también influencia de los ácidos grasos sobre las cifras postprandiales de glucosa y lípidos. El objetivo del presente estudio es determinar si los niveles postprandiales de glucosa y lípidos difieren entre un desayuno con ácidos grasos saturados vs ácidos grasos insaturados en pacientes con diabetes mellitus tipo 2 (DMT2). Métodos: Es un estudio de tipo cuasi experimental con diseño cruzado, en el cual se realizó eva-luación antropométrica (peso, talla, IMC, circunferencia de cintura, porcentaje de grasa y músculo) y evaluación bioquímica preprandial y postprandial (glucosa, colesterol total, colesterol HLD, colesterol LDL, triglicéridos) en 30 pacientes adultos con diabetes tipo 2, a los cuales se les administró dos tipos de desayunos isocalóricos, el uno con aporte de ácidos grasos saturados 99,3 Kcal y el otro de ácidos grasos insaturados 113,4 Kcal. Resultados: la ingesta de ácidos grasos saturados produce una mayor elevación de las cifras postpran-diales de glucosa (p=0,01) y no de los lípidos postprandiales. Los ácidos grasos insaturados, permitieron llegar al objetivo de glucosa postprandial recomendada (<140mg/dL), en un mayor porcentaje de pacientes (p=0,02). Conclusiones: el consumo de un desayuno alto en ácidos grasos insaturados permitió un mejor control de las cifras postprandiales de glucosa en comparación con el consumo de un desayuno alto en ácidos grasos saturados. No existió diferencia en las cifras de lípidos posprandiales en los dos tipos de desayuno


Background and objective: type 2 diabetes mellitus is a disease that has been increasing rapidly in low- and middle-income countries, affecting a large population group. Being the carbohydrates, the main determinant of the postprandial glucose elevation, it has been evidenced that there is also an influence of the fatty acids on the postprandial values of glucose and lipids. The aim of the present study is to compare the postprandial levels of glucose and lipids between a breakfast high in saturated fatty acids vs unsaturated fatty acids in patients with T2DM. Materials and methods: it is an cuasi experimental crossover study. We did an anthropometric evaluation (weight, height, BMI, waist circumference, percentage of fat and muscle) and preprandial and postprandial biochemical evaluation (glucose, total cholesterol, HLD cholesterol, LDL cholesterol, Triglycerides) in 30 adults patients with type 2 diabetes, to determine the influence of fatty acids in two different breakfasts with the same caloric load, but each with a different contribution of saturated 99.3 Kcal or unsaturated fatty acids 113.4 Kcal. Results: the intake of a breakfast high in saturated fatty acids produced a higher increase in postprandial glucose values (p = 0.01) and not in postprandial lipids. Unsaturated fatty acids permit attain the goal of postprandial glucose (<140mg /dL) (p = 0.02) in most patients. Conclusions: The intake of high unsaturated fatty acids breakfast allowed a better postprandial glucose control, in comparison to the consumption of a breakfast high in saturated fatty acids. We did not find any difference in postprandial lipids with both types of breakfast.

Using technical procedures, the fatty acid (FA) profile and solid fat content (SFC) of the Peruvian cultivar cacao beans CCN 51 and ICS 6 and the "optimal chocolate", obtained from the mixture of the first two, were determined to assess their quality. These cacao beans were found to have important nutritional values. The FA profile of the cacao beans were similar (p>0.05); however, in the FA profile, the 'optimal chocolate' had significant differences (p≤0.05) in terms of palmitic, arachidic and linolenic acid. The n6:n3 ratio for "optimal chocolate" was 12.0 ± 1.7. Cacao beans had the same SFC, and SFC was highly temperature dependent, as determined using a mathematical model for chocolate. The SFC of chocolate refers to hard cacao butter content at temperatures between 20 and 25°C, and solid fat was heat resistant from 25 to 30°C, which is considered valuable in trade chocolate production. The quality-related properties of these lipid fractions imparted nutritional and physical aspects to the optimal dark chocolate for human consumption.


La composición de ácidos grasos (CAG) y el contenido de sólidos grasos (CSG), de la fracción lipídica de los cultivares peruanos de cacao CCN 51 e ICS 6 así como del "chocolate óptimo", obtenido de las mezclas de las primeras dos, fueron determinados por técnicas analíticas para conocer su calidad. Estas variedades tuvieron valores nutricionales importantes. La CAG de los granos de cacao fueron similares, sin embargo la CAG del "Chocolate óptimo" tuvo diferencias significativas (p<0,05) para los acidos grasos palmitico, araquidico y linoleico. El ratio n6:n3 fue de 12,0 ± 1,7. El CSG de los granos de cacao fueron los mismos y tuvo una fuerte dependencia con la temperatura, también se definió un modelo matematico para el chocolate. El CSG le confiere al chocolate una consistencia dura a temperaturas de 20 a 25°C y resistentes al calor de 25 a 30°C, siendo tales propiedades una ventaja en la comercialización de chocolates. La calidad de estas fracciones lipidicas tuvieron aspectos nutricionales y fisicos en el chocolate oscuro para consumo humano.

ABSTRACT Polyunsaturated fatty acids oxidize easily due to their chemical structure, causing a reduction of their nutritional properties. Nanostructured systems may be an alternative to protect fatty acids against oxidation, improving solubility and stability. Consequently, nutritional value of food is maintained as well as the sensory characteristics (color, flavor, texture, and aroma) when fatty acids are added to food products. The present study is a narrative review to introduce the potential benefits of omega-3 unsaturated fatty acids nanoparticles incorporated in food products. The literature review includes publications in English and Portuguese issued between March 1985 and March 2019, in PubMed, ScienceDirect and Web of Science databases. Manual searches were conducted in the articles references lists of the articles included to identify other relevant studies. There were studies that evaluated the stability of fatty acids in food products such as bread, fruit juice, milk, yogurt, and meat. In this study, the most used nanostructured systems for the incorporation of fatty acids were the nanocapsules and the nanoliposomes. Currently, the nanostructured system demonstrates a potential to improve protection of polyunsaturated fatty acids against oxidization and thermal degradation. In this way, they maintain their functional properties and their bioavailability increases and therapeutic efficacy and sensory properties are improved. There are several methodologies being tested, which makes it difficult to identify the most efficient formulation to protect fatty acids. Nanostructured systems seem to be the best alternative to protect polyunsatured fatty acids from oxidization. The encapsulation efficiency, particle's size and type are relevant factors to be considered to evaluate oxidization. In conclusion, the review showed that currently it is impossible to determine the most efficient methodology. Besides, nanoformulations should follow international guidelines to present more standardized and therefore more efficient particles.


RESUMO Os ácidos graxos poli-insaturados são facilmente oxidados devido à sua estrutura química, o que acarreta a diminuição de suas propriedades nutricionais. Nesse contexto, o sistema nanoestruturado pode ser uma alternativa para protegê-los contra a oxidação, melhorando a solubilidade e estabilidade. Consequentemente, quando são adicionados a produtos alimentares, o valor nutricional é mantido, bem como as características sensoriais (cor, sabor, textura e aroma). O presente estudo é uma revisão narrativa para apresentar os potenciais benefícios das nanopartículas com ácidos graxos insaturados da classe ômega-3 incorporados a produtos alimentícios. A literatura consultada incluiu publicações em inglês e em português, considerando o período entre março de 1985 e março de 2019, utilizando as bases de dados PubMed, ScienceDirect e Web of Science. Foram realizadas buscas manuais nas referências dos artigos incluídos, a fim de identificar outros estudos relevantes. Foram encontradas pesquisas que avaliaram a estabilidade dos ácidos graxos em produtos alimentícios, como pães, suco de fruta, leite, iogurte e carne. Neste estudo, as nanoestruturas mais utilizadas para a incorporação dos ácidos graxos foram as nanocápsulas e os nanolipossomas. Atualmente, o sistema nanoestruturado demonstra um potencial para melhorar a proteção desses ácidos poli-insaturados contra a oxidação e degradação térmica. Dessa forma, eles mantêm suas propriedades funcionais, aumenta-se sua biodisponibilidade e há melhora da eficácia terapêutica e das propriedades sensoriais. Existem diversas metodologias sendo testadas, o que dificulta a identificação de qual seria a formulação mais eficiente nessa proteção, mas os sistemas nanoestruturados parecem ser a melhor alternativa para proteger os ácidos graxos insaturados da oxidação. A eficiência de encapsulação bem como o tamanho e o tipo de partícula são fatores importantes a considerar na avaliação da oxidação. Em conclusão, a revisão demonstrou que atualmente a metodologia mais eficiente não é possível de ser identificada. Ademais, as nanoformulações devem seguir diretrizes do protocolo internacional para apresentar partículas mais padronizadas e, assim, eficientes.

OBJECTIVE: To discover an efficient strategy for a conversion of the antibacterial activity into an antitumor activity. METHODS: Pharmacophore and scaffold hopping-based rational drug design principles, the title fluoroquinolone C-3 thiazolotriazole unsaturated ketones (6a-6l) were designed and synthesized with a S-triazole ring and α,β-unsaturated ketone, respectively, as an isostere and fused modified group from ofloxacin (1), and their structures were characterized by elemental analysis and spectral data, and the in vitro antitumor activity against the tested tumor cell lines was evaluated by a MTT assay. RESULTS: Twelve new title compounds were synthesized, and exhibited more significant potency than parent 1. The title compounds with fluorophenyl or O-methoxyphenyl displayed comparable activity to comparasion doxorubicin. CONCLUSION: A fused heterocyclic unsaturated ketone skeleton as an isostere of the C-3 carboxylic acid group are shown to be an alternative route for further design of lead antitumor fluoroquinolone.

ABSTRACT The need to obtain nutritious foods from new sources and lower waste in industry has created a high interest in studying different parts of plants or foods that today are considered waste, but could be considered by-products with high nutritional value with potential use in human diets. Pumpkin seeds are commonly considered as waste but they have a high content of fatty and amino acids, which when used as a by-product or ingredient can add value to food products. The aim of this work was to perform a wide review of the nutritional and functional properties of Cucurbita maxima seeds and their potential medicinal influence.


RESUMEN La necesidad de obtener alimentos nutritivos de nuevas fuentes y menores desperdicios en la industria ha generado un gran interés en el estudio de diferentes partes de plantas o alimentos que hoy en día se consideran desechos, pero que podrían considerarse subproductos con alto valor nutricional y uso potencial en alimentación humana. Las semillas de calabaza se consideran comúnmente como desechos, pero tienen un alto e importante contenido de ácidos grasos y aminoácidos, que cuando se utilizan como subproducto o ingrediente pueden aportar un alto valor agregado a los productos alimenticios. El objetivo de este trabajo es realizar una amplia revisión de las propiedades nutricionales y funcionales de las semillas de Cucurbita maxima y su potencial influencia medicinal asociada a ellas.

ABSTRACT

The purpose of this study was to determine and compare fat composition and chemical properties of fish fillets of selected warm-water fish obtained from Straits of Malacca. A cold water fish, namely salmon was used for comparison. Moisture content, crude fat, fatty acids composition and chemical characteristics of fish fillets of Yellowstripe scad, Japanese threadfin bream and salmon were determined. Japanese threadfin bream fillet had highest moisture and crude fat contents, followed by fillets of Yellowstripe scad and salmon. A significantly strong and negative correlation was found between moisture and crude fat contents of these fish fillets. Fillets of Japanese threadfin bream and Yellowstripe scad also had higher total saturated fatty acids than total unsaturated fatty acids. Although salmon fillet had lowest percentage of saturated fatty acids, it had highest monounsaturated fatty acids and polyunsaturated fatty acids (PUFA) compared with the two warm-water fish. Palmitic acid and oleic acid were the major fatty acids in the fish fillets. Chemical properties of the oils extracted from the warm-water fish fillets were varied compared to salmon. The selected warm-water fish fillets offer favorable fatty acids composition and chemical properties, which can potentially be used as good sources of PUFA.


O objetivo deste estudo foi determinar e comparar a composição gordurosa e as propriedades químicas de filés de peixes selecionados de água quente obtidos no Estreito de Malaca. Um peixe de água fria, o salmão, foi usado para comparação. Foram determinados o teor de umidade, a gordura bruta, a composição de ácidos graxos e as características químicas dos filés de yellowstripe scad, sargo japonês e salmão. Os filés de sargo japonês apresentaram maior teor de umidade e de gordura bruta, seguidos por filés de yellowstripe scad e de salmão. Foi encontrada uma correlação significativamente forte e negativa entre a umidade e o teor de gordura bruta desses filés de peixe. Filés de sargo japonês e de yellowstripe scad também apresentaram ácidos graxos saturados totais mais altos do que os ácidos graxos insaturados totais. Embora o filé de salmão tenha menor porcentagem de ácidos graxos saturados, ele apresentou os maiores ácidos graxos monoinsaturados e ácidos graxos poliinsaturados (PUFA em comparação com os dois peixes de água quente. O ácido palmítico e o ácido oleico foram os principais ácidos graxos dos filés de peixe. As propriedades químicas dos óleos extraídos dos filés de peixe de água quente foram variadas em comparação ao salmão. Os filés de peixe de água quente selecionados oferecem composição favorável de ácidos graxos e propriedades químicas, que podem ser potencialmente usadas como boas fontes de PUFAs.

Background: Type 2 diabetes is the result of complex interplay between genetic and environmental factors. Diabetes epidemic is largely due to unhealthy diet and lifestyles. Dietary composition affects both its development and complications because fatty acids influence glucose metabolism by altering cell membrane function, enzyme activity, insulin signalling, and gene expression. Therefore, replacing saturated fats and trans-fatty acids with unsaturated fats has beneficial effects on insulin sensitivity and in reducing the risks of type 2 diabetes. Research has shown that if people with diabetes, together with the health care workers manage diabetes well, that the risk of complications will be greatly reduced or prevented or delayed. This study focused on assessing the prevalence and causes of type 2 diabetes, types of complications patients presented, and the likely preventive interventions given to them in University Teaching Hospital Orlu.Methods: A retrospective study that carried out extensive review of hospital records of adults admitted and treated for type 2 diabetes mellitus from 2015 to 2017 in University Teaching Hospital was done. The review of the adult patients' records lasted for three working weeks. In the review, summaries of causes of diabetes, types of complications and preventive measures health workers provided to the patients were documented.Results: Finding showed that 79 adults made up of 43(54.4%) males and 36(45.6%) female between 41-77 years were admitted and treated for various causes of type 2 diabetes between 2015 and 2017. Most of the patients treated were civil and public servants. The main cause of diabetes mellitus 51(65%) among the patients studied was physical inactivity due to sedentary work. The common complications presented were hypertension, diabetic ulcer, depression and obesity. Health talks and group counselling were the interventions provided. These interventions concentrated on types of healthy dietary intakes and lifestyles the patients should adopt.Conclusions: Regular and sustained health care services that would emphasize healthy diet and lifestyle modifications to reduce the risk factors of type 2 diabetes mellitus should be provided to all patients treated in this hospital. Therefore, obese diabetic patients should be encouraged to engage in physical exercises so as to maintain healthy body weight which is a cornerstone for prevention of type 2 diabetes mellitus.

To expand an efficient strategy for the conversion of antibacterial activity of fluoroquinolones into an antitumor activity, sixteen new compounds, 1-cyclopropyl-6- fluoro-7-(4-methyl-piperazin-1-yl)-3-(5-arylidene-thiazol-4(5H)-one-2-yl)-quinolon-4(1H)-ones (7a-7p), were designed and synthesized with a thiazolone ring and an arylidene moiety as an isostere and modified group, respectively, from ciprofloxacin. Their structures were characterized by elemental analysis and spectral data. The in vitro antitumor activity of the synthesized compounds were measured using Hep-3B, Capan-1 and HL60 cell lines and were found to be more potent than ciprofloxacin. Meanwhile, the SAR revealed that the halogenated phenyl compounds such as fluorophenyl (7h, 7i), chlorophenyl (7j, 7k) or bromophenyl compounds (7l, 7m), and aromatic heterocyclic substitution such as furyl (6n) or pyridyl compounds (6o, 6p) displayed better activity than the control compounds, especially the IC50 values of pyridyl compounds 6o and 6p against Capan-1 cell growth was comparable to doxorubicin. Thus, an arylidene-modified thiazolone scaffold as the replacement of the C-3 carboxylic acid group appears to be an alternative route for an improved antitumor activity of fluoroquinolones.

ABSTRACT

In the present study, two new acetylene conjugate compounds, dibutyl (2Z, 6Z)-octa-2, 6-dien-4-yne dioate (1), and dibutyl (2E, 6E)- octa-2, 6-dien-4-yne dioate (2), were isolated from the dry stem leaves of Viscum album, along with nine known compounds (3 - 11). Their structures were confirmed on the basis of spectroscopic data. Compounds 1 and 8 showed antioxidant activity against xanthine oxidase (XOD) and 1,1-diphenyl-2-picrylhydrazyl radical 2,2-diphenyl-1-(2,4,6-trinitrophenyl) hydroxyl (DPPH), with the IC of 1.22 and 1.33 μmol·L, and the SC of 4.34 and 8.22 μmol·L, respectively.
